Abstract

GLRaV-2 belonging to the Closterovirus genus of the family Closteroviridae is one of the 5 virus species responsible for leafroll disease of grapevine. GLRaV-2 species encompasses 6 molecular variants exhibiting up to 30% nucleotide divergence. Development of a GLRaV-2 detection test by real-time RTPCR has required design of specific and universal primers towards these different variants. This realtime RT-PCR detection method was validated with a panel of 85 vine samples collected worldwide, 10 of them being negative with ELISA method but readily detected with this new method. Its detection sensivity was increased 125 times compared to a conventional single-tube RT-PCR. Use of this method for detection of GLRaV-2 in rootstocks has shown its higher sensitivity and reliability especially for root tissues.
