Using 281 fb^\ensuremath-1 of data from the Belle experiment recorded at or near the \ensuremathΥ(4S) resonance, we have measured the rates of the ``wrong-sign'' decays D0\ensuremath→K+\ensuremathπ^\ensuremath-\ensuremathπ0 and D0\ensuremath→K+\ensuremathπ^\ensuremath-\ensuremathπ+\ensuremathπ^\ensuremath- relative to those of the Cabibbo-favored decays D0\ensuremath→K^\ensuremath-\ensuremathπ+\ensuremathπ0 and D0\ensuremath→K^\ensuremath-\ensuremathπ+\ensuremathπ+\ensuremathπ^\ensuremath-. These wrong-sign decays proceed via a doubly Cabibbo-suppressed amplitude or via D0\mathrm\text\ensuremath-D0 mixing; the latter has not yet been observed. We obtain RWS(K\ensuremathπ\ensuremathπ0)=[0.229\ifmmode±\else\textpm\fi0.015(stat)_\ensuremath-0.009+0.013(syst)]% and RWS(K3\ensuremathπ)=[0.320\ifmmode±\else\textpm\fi0.018(stat)_\ensuremath-0.013+0.018(syst)]%. The CP asymmetries are measured to be \ensuremath-0.006\ifmmode±\else\textpm\fi0.053 and \ensuremath-0.018\ifmmode±\else\textpm\fi0.044 for the K+\ensuremathπ^\ensuremath-\ensuremathπ0 and K+\ensuremathπ^\ensuremath-\ensuremathπ+\ensuremathπ^\ensuremath- final states, respectively.
