Plums are an important part of the fruit-growing industry. This study was focused on the evaluation of tolerant and hypersensitive cultivars of plum (Prunus domestica L.) exposed to natural infection pressure of Plum pox virus (PPV) under the conditions of an experimental orchard in Eastern Bohemia. Ten genotypes were monitored, including traditional tolerant varieties (‘Čačanská lepotica’, ‘Durancie’, ‘Gabrovská’, ‘Haganta’, ‘Tophit’) and hypersensitive varieties (‘Jojo’, ‘Jofela’, ‘Joganta’), along with hybrids created by breeding of variety ‘Jojo’ (HL 900045, HL 900204). Phenological and production traits, flower and fruit set, fruit quality, and sugar content were evaluated in the years 2021–2025. The results show that cultivars with a hypersensitive reaction have comparable production levels and fruit quality to tolerant genotypes. The new hybrids are characterized by specific ripening times, which increase the variability of the cultivation portfolio. In conclusion, all studied cultivars show potential for long-term cultivation in the region, and hypersensitive genotypes represent a suitable complement to tolerant cultivars due to their defence strategy and production stability.
