Abstract

A rapid method of solid phase extraction (SPE) has been developed for the stability of melatonin (MEL) in commercial purple grape juice (PGJ) and two varieties of commercial red fruit juices (RFJ and RFJ+Bb); the solid phase and the solvent employed have been Strata-X and methanol. Therefore, the determination of melatonin levels and melatonin stability in those juices is very interesting information to study and to compare it level with others kinds of juices. The linear range of the melatonin standard curve was from 0.7 to 10 ng. mL-1 (R2= 0.99). Precision for inter-day and intra-day assay showed acceptable results for quantitative assay for melatonin standard solutions and melatonin in spiked sample of grape juice and red fruit juices. The recoveries were found to be within the range of 101 and 103% for the PGJ, 100% for the RFJ, and within the range of 74 and 102% for RFJ+Bb. The results obtained indicate that the solid phase extraction could provide excellent reproducibility of melatonin in juices indicated, shown in this way prior to the study design process of this compound extraction for others varieties of juices.
