Plant tissue techniques have been used for decades for commercial plant multiplication and research purposes. For some species not easily propagated under in vitro conditions, efficient micropropagation protocols are still missing. With the rapid development of genetic modification techniques, a need for functional micropropagation protocols for recalcitrant, but also agriculturally important plants are rising. Sweet cherry (Prunus avium L.) belongs to this group by exhibiting distinctive genotypic variability when micropropagated on culture media. The causes of this recalcitrance may be related to physiological state of the plant, the composition of the culture media or defensive reactions of the plant. The present study compares the influence of two culture media (BAP 200 and QL-1) on the survival rates of explants of six genetically valuable sweet cherry cultivars under in vitro culture conditions. Results show clear cultivar specificity for micropropagation on culture media (e.g. genotype HL 10072) and persistent recalcitrance of certain perspective varieties (e.g. ʽKordiaʼ), which hinders their use for genetic modifications.
